Case study
A repeat customer made a request for us to fully develop and validate an assay to measure MAP4K5 (KHS1) activity. In addition to the assay development, we cryptate labeled the anti-phospho antibody and procured a biotinylated substrate to allow for the use of Streptavidin-XL665 for detection.
The assay was to be utilized in high-throughput screening to identify modulators of the enzyme's activity. With this in mind, a plan for assay development was put in place and communicated to the customer. This included the kinetic characterization of the enzyme with respect to rate, Km of ATP and substrate as well as inhibitor profiling. Further, the stability of the reagents diluted over time and the statistical robustness of the data was assessed to ensure that the assay would be suitable for high-throughput screening. Following the complete validation, a report was generated and the assay was transferred to the customer.

Independent IC50 determinations for staurosporine demonstrated that the assay was able to give reproducible results, falling in the literature reported range of activity for the inhibitor. Further, the IC50 value generated with enzyme diluted over three hours was in line with those values generated using freshly prepared material. This is indicative of the stability and robustness of the assay.

Statistical data for various experiments indicate that the assay is reproducible as well as statistically robust. The S/B and z' ratios indicate that the screen should perform well in an automation setting.
