SNAP substrate features:
  • non-permeating to membrane cells, and specifically label only the GPCR expressed at the cell surface.
  • small molecules (< 2 kDa) that do not affect the specific ligand binding.
  • chemically inert for other proteins, reducing non-specific binding.

How does the SNAP-tag substrate work?

SNAP- tag is a suicide enzyme, and can be easily fused to the N- terminal position of GPCR. SNAP-tag can then be specifically and covalently labelled with a SNAP-tag specific substrate, benzyl guanine and benzyl cytosine respectively. This substrate is labelled with terbium cryptate (Lumi4-Tb).
