ApoA1 and ApoB assays
Apolipoprotein A1 (ApoA1) and Apolipoprotein B (ApoB) belong to the lipoprotein family, which plays important roles in lipid metabolism. While ApoA1 is the main component of HDL (high-density lipoprotein, or "good cholesterol"), ApoB is the main component of LDL and VLDL (low and very-low density lipoprotein, or "bad cholesterol").
The transcription of ApoA1 and ApoB genes is up-regulated by numerous transcription factors including PPAR,RXR, HNF4alpha, and LXR.
The levels of ApoA1 and ApoB are important markers for evaluating the risk of arteriosclerosis.

Assay Principle:
The Apo-A1 and Apo-B assays can be run with cell supernatants or whole cells using the protocols shown here. Apolipoproteins are measured using sandwich immunoassays involving two monoclonal antibodies: anti-Apo MAb1 labeled with Eu-Cryptate and anti-Apo MAb2 labeled with d2. These antibodies may be pre-mixed and added in a single dispensing step to further streamline the protocol.
The assay can be run either under a cell-supernatant assay protocol (two-plate), or be further streamlined to a cell-based assay protocol (one-step).
