Amyloid β1-40 HTRF® Assay
Amyloid β1-40 and β1-42 peptides are involved in the appearance of Alzheimer's disease symptoms. The production of these two amyloid peptides is regulated by the activity of 2 enzymes, β and γ secretase, which cleave the amyloid precursor protein. Cisbio Bioassays developed an assay for the measurement of amyloid β1-40 peptides based on our proprietary HTRF® technology. This assay can be rapidly performed, easily miniaturized and is highly amenable to automation.

Assay Principle:
The HTRF® Amyloid β1-40 HTRF assay is a sandwich immunoassay which uses two monoclonal antibodies which recognize different epitopes within the β1-40 peptide. One antibody is labeled with Eu3+-Cryptate and the second with XL-665. The assay can be used with whole cells or cell supernatants.
Assay Protocol:
The Amyloid β1-40 peptide assay can be performed in a single microplate, from cell stimulation through HTRF® signal detection. The quantification process involves the addition of the two specific antibodies-conjugates which can be pre-mixed allowing for a single reagent addition protocol. This simplified protocol can be miniaturized and is suitable for HTS.
