Antibody screening on cell surface targets
Tag-lite® is a cellular screening platform that enables the investigation of antibody binding to cell surface proteins, such as GPCRs and RTKs. The surface protein of interest is expressed as a fusion protein with a SNAP-tag®. Upon protein expression and transport to the cell surface, this tag can be covalently labeled to an HTRF® dye using a fluorophore-bearing substrate (1). Once the process is completed, the labeled cells are ready to be used in HTRF Tag-lite® assays. The technology is very flexible, and as illustrated here, can be optimized with 3 main assay formats.
